Cell culture equipment and introductory tutorial preparation room equipment:

Single distilled water distiller, double distilled water distiller, acid tank, oven, pressure cooker, storage cabinet (placed unsterilized items), storage regulations (placed with sterilized items), packaging table. Equipment for the dispensing chamber: torque balance and electronic balance (for weighing drugs), pH meter (for measuring the pH of the culture solution), magnetic stirrer (for mixing the solution in the dispensing chamber).

Equipment in the cultural room:

Liquid nitrogen tank, storage cabinet (storage), fluorescent and UV lamps, air purification system, low temperature refrigerator (80 ° C), air conditioning, carbon dioxide gas cylinder, side (write experimental record). Must be placed between sterile equipment: centrifugation (collection), ultra-clean bench, inverted microscope, CO2 incubator (culture culture), water bath, trioxo, 4 °C refrigerator (serum and liquid culture).

Aseptic operation

Sterilize in a sterile room:

Clean the sterile room regularly: once a week, mopping the floor with tap water, wiping the table, cleaning the workbench, etc., then clean the room with a 3 ‰ or New Street or 0.5% peracetic acid.

2. Sterilization of CO2 incubator (culture chamber): first wipe with 3‰xingjieer, then wipe with 75% alcohol or 0.5% peracetic acid, then irradiate with UV light

3. Pre-experimental sterilization: Each time the UV lamp, oxygen sterilizer and air purifier system are turned on for 20-30 minutes

4. Post-experimental sterilization: Wipe the ultra-clean platform of the inverted microscope with 75% alcohol (3‰xinjiermian), the side platform and the load carrier platform.

Aseptic preparation of the experimenter:

Wash your hands with soap.

2. Put on the isolation suit, wear the isolation cap, mask and slippers

Wipe your hands with a 75% alcohol tampon.

Demonstration of aseptic operation:

1. All alcohol, PBS, medium and trypsin bottles loaded into the ultra clean bench should be wiped with 75% alcohol on the outer surface of the bottle.

2. Approach the alcohol lamp flame operation.

3. Must be sterilized before using the appliance

Continue to use utensils (such as caps and droppers) at high places and use them on the counter.

5. All kinds of operations should be close to the alcohol lamp, the movement should be light and accurate, and should not be touched at will. If the straw does not touch the waste tank.

6. Replace the straw when absorbing two or more liquids to prevent cross-contamination.
